    Effects of Substrate on the Electricity Generation Characteristics of Microbial Fuel Cell

    A reaction equipment of microbial fuel cell (MFC) with “H” structure was designed and fabricated.The sludge collected from the aeration tan in Datianwan Sewage Treatment Plant in Jishou was used to inoculate electricigens.The effects of substrate species and concentration on the electricity generation of MFC in view of start-up time,time for obtaining stable open circuit voltage,electromotive force,internal resistance and power density were investigated.Experimental results showed that the substrate species and concentration greatly influenced the electricity generation of MFC probably due to their difference in structure.It was found that the white dextrin-fed MFC could be started up in shorter time with smaller internal resistance,smaller electromotive force and smaller power density.Under the same conditions,the soluble starch-fed MFC had larger electromotive force,larger power density,bigger internal resistance and longer time to be started-up.The results also indicated that an increase in the substrate concentration increased the time for reaching constant OCV at low concentration of 2~6 g/L for soluble starch and 2~8 g/L for white dextrin.While an opposite trend was observed at high concentration of 8~20 g/L for soluble starch and 10~20 g/L for white dextrin attributed to the substrate inhibition effect.

    Separation and Characterization of Catechin in man-Made Green Tea,the Fruits and Leaves of Berchemia Floribunda with Macroporous Resin and HPLC

    In this study,the extraction process of catechins was investigated with a certain concentration of ethanol as solvent from man-made green tea,fruits and leaves of Berchemia floribunda,using the method of waterbath.After the fat soluble pigment was removed by petroleum ether from extracting solution,the catechins was purified by porous resin adsorption method.The static adsorption and desorption tests of catechin with macroporous resin showed that D941 was the optimal resin for purification and the pure water was the optimum eluant.Purified liquid of fruit,leaves and home-made green tea extracts were detected by HPLC.The catechins was separated on PC18 chromatographic column (4.6×250 mm,5 μm) at 35 ℃,with nitrile-0.4% phosphoric acid (13∶87) as mobile phase at a flow rate of 1 mL/min,and detected at the wavelength of 279 nm (UV scan).The results showed that the catechin content of fruit was the lowest,while the content of man-made green tea was significantly higher than that of both the fruit and leaves through the optimized extraction processes.

    Antitoxic Effect of Extracts of Tripterospemum Cordatum Against  Deinagkistrodon Acutus Snake Venom

    The antitoxic effect of Tripterospemum cordatum aqueous extracts against Deinagkistrodon acutus snake venom is researched.The BACA/C mice were divided into five group according to the dosage of Tripterospemum cordatum aqueous extracts applied:low dosage,middle dosage,high dosage,blank group and model group,each group of health conditions being observed after 7-days intra-gastric administration.For the last time of administration,each group was intraperitoneally injected with Agkistrodon acutus snake venom after one hour later,then the mice’s poisoning performance was observed and the death rate was calculated,and each group of mice’s tissue bleeding,death and ulceration compared.The results showed that the aqueous extract of Tripterospemum cordatum at the low dosage,middle dosage and high dosage all reduced the mortality caused by Agkistrodon acutus snake venom poisoning and relieved some symptoms;with the increase of the dosage the symptoms decreased significantly,and the protection effect on the tissue was especially apparent.It can be concluded that the aqueous extract of Tripterospemum cordatum has antitoxic effect against Agkistrodon acutus snake venom and decrease the toxicity.

    Temporal Scaling Characteristics of Correlation of O3 and NO2 for Urban Area of Chengdu  in  Basin Climate

    The correlations between ground-level NO2 and O3 concentration are analyzed by the method of detrended cross-correlation analysis,based on the observational data in the urban area of Chengdu (capital of Sichuan Provincial,China) from July 1 to August 31,2013 and from January 1 to February 28,2014.The results show that the correlation between NO2 and O3 concentration are characterized by long-term persistence.Furthermore,a comparative study of the correlation of NO2 and O3 in the summer and winter is made,and the reason for the differences are analyzed.It shows that there were significant differences in meteorological conditions on the different time scale of the interaction between NO2 and O3.It was found that the differences are due to solar radiation,sunshine hours and temperature.The results help to deepen the understanding the pollution characteristic of O3 in Chengdu.

    Literatures abroad on energy consumption and carbon emissions of different building structures are reviewed,with focus on the research progress and research results about the energy consumption and carbon emissions of wooden structures,concrete structures and steel structures.The article draws the conclusion that the energy consumption and carbon emissions of wooden structures is lower than those of the concrete and steel ones;wooden structure has a carbon storage function and waste wood burning can substitute fossil fuels;at construction sites,concrete structures consume more energy than steel structures do;under the same boundary conditions,concrete structures have lower implied energy and implied carbon than steel structures;but if the steel recovery is considered,energy consumption and carbon emissions of both the structures are appropriate.This conclusion can provide reference for the domestic study on energy consumption and carbon emissions of different building structures in China.

    Diversity Investigation of Vascular Plant Species in Donghe National Wetland Park

    The diversity of vascular plant species in Donghe National Wetland Park is described by field surveying.The results show that there are 1485 species of vascular plant in Donghe National Wetland Park,belonging to 766 genera and 179 families.Among them,there are 285 Wetland plants (including aquatic plants),belonging to 186 genera and 73 families,accounting for 40.78%、24.28% and 19.19% of the total amount of families,genera and species in Donghe National Wetland Park,respectively.The geographic distribution is complex for 11 types of family distribution and 15 types of genera distribution.The flora in genera displayed significant tropical feature,with 46.25% (including 327 genera) of tropical elements and 41.00% (including 288 genera) of temperate elements.The corresponding conservation strategies were proposed,aiming to provide reference for the biodiversity conservation of National Wetland Park.

    The antimicrobial activity of the fermentation brothes of 294 halophilic and halotolerant bacteria,isolated from ordinary non-saline soils collected from the Dehang Canyon (28°15′~28°43′ N,109°30′~109°45′ E) in Hunan Province,China,was screened using the agar diffusion method employing 8 indicator microorganisms (three Gram-positive bacteria,three Gram-negative bacteria and two fungi).The results showed that,out of 294 strains tested,109 were positive in antimicrobial activity (37.1%),in which 5 strains exhibited strong antimicrobial activity.Then these 5 strains were submitted for 16S rRNA gene amplification and phylogenetic analysis after phenotypic characterization including morphological,biochemical and physiological tests.The results suggested that these 5 strains were members of 4 genera in 3 families (Bacillaceae,Halomonadaceae,Nocardiopsaceae) of 3 phyla (Actinobacteria,Firmicutes,Proteobacteria):strain JSM 102030 belonged to the genus Halobacillus (Firmicutes,Bacillaceae),being most closely to the type strain of Halobacillus trueperi (sequence similarity 99.1%);strains 102052 and JSM 102054 were members of the genus Halomonas (Proteobacteria,Halomonadaceae),being related most closely to the type strain of Halomonas alkaliphila (98.3%);strains JSM 102066 strain JSM 2155017 belonged to the genus Nocardiopsis (Actinobacteria,Nocardiopsaceae),exhibiting 99.3 % 16S rRNA gene sequence similarity to Nocardiopsis prasina DSM43845T;strain JSM 102082 belonged to the genus Oceanobacillus (Firmicutes,Bacillaceae),exhibiting 99.6% 16S rRNA gene sequence similarity to Oceanobacillus kimchii X50T.

    Isolation and Plant Growth-Promoting Ability of ACC Deaminase-Containing Bacteria from Rhizosphere of  Gynostemma Pentaphyllum Makino

    A total of 9 bacterial strains were isolated from rhizosphere of Gynostemma pentaphyllum through enrichment on 1-aminocyclopropane-1-carboxylate (ACC) as a sole nitrogen source.ACC deaminase assay showed that all isolates differed in their potential for ACC deaminse activity,among which 3 strains were positive for siderosphere by chrome azurol S (CAS) assay.Inoculation of rice seedlings with 7 ACC deaminase-containing strains had significant effect on root elongation and root fresh weight,among which the strain JDG127 caused 1.6-fold increase in root length and root fresh weight over uninoculated control.Based on the physiological and biochemical characteristics in combination with 16S rRNA gene sequences analysis,the 9 ACC deaminase- containing strains were identified as Stenotrophomonas (1 strain),Paenibacillus (1 strain),Bacillus (1 strain),Enterobacter (2 strains) and Acinetobacter (4 strains).The results indicated that the siderosphere producing ability of rhizobacteria may be correlated with their plant growth-promoting ability when compared to their ACC-deaminase activity.

    The culturable bacteria in the air samples from a liquor starter-making workshop in Jiugui Liquor Co.Ltd.,China were isolated by using the natural precipitation method and culture-dependent method.The amylase-producing ability of the strains was investigated by using the plate hydrolysis spot method.The results showed that 16 out of 65 (24.6%) strains tested exhibited amylase-producing activity,in which 4 strains (JSM 2145008,JSM 2155001,JSM 2155017,JSM 2175012) could relatively strongly produce amylase.Subsequently,these 4 strains were investigated by means of a phylogenetic analysis based on 16S rRNA gene sequence comparisons.The results showed that these 4 strains were the members of 3 families (Bacillaceae,Deinococcaceae,Micrococcaceae) in 3 phylogenetic group/phyla (Actinobacteria,Deinococcus-Thermus,Firmicutes):strain JSM 2145008 belonged to the genus Arthrobacter (Actinobacteria,Micrococcaceae),being most closely to the type strain of Arthrobacter oxydans (sequence similarity 99.8%);strains JSM 2155001 and JSM 2175012 were the members of the genus Deinococcus (Deinococcus-Thermus,Deinococcaceae),being related most closely to the type strains of Deinococcus daejeonensis (99.6%) and Deinococcus yunweiensis (99.9%),respectively;strain JSM 2155017 belonged to the genus Bacillus (Firmicutes,Bacillaceae),exhibiting 100% 16S rRNA gene sequence similarity to the type strain of Bacillus stratosphericu.In conclusion,there is abundant bacterial diversity,as well as a number of amylase-producing strains in the air samples from a liquor starter-making workshop in Jiugui Liquor Co. Ltd.

    The factors affecting the ISSR-PCR amplification on Ficus pumila L. var. awkeotsang such as dosage of template DNA,Taq DNA polymerase,dNTPs,primers and extension time and cycle times were optimized through the combination of single factor and orthogonal design experiment.The results showed that the optimal reaction system for ISSR-PCR of Ficus pumila were as follows:In the 20 μL reaction volume,1ng template DNA,1.0U Taq DNA polymerase,0.4 μmol/L primers,0.18 mmol/L dNTPs.The optimal PCR amplification process was:4 minutes at 94 ℃ for predenaturation,then followed by 35 cycles,each with 45 seconds at 94 ℃ for denaturation,45 seconds at 53 ℃ for annealing,2 at 72 ℃ for extension,finally extension at 72 ℃ for 7 minutes and holding the samples at 4 ℃.The system was applied in the amplification of 14 varieties of indicated the suitability and stability of the system.

    Investigation and Analysis of Yellow Flower Plants Cultivation in Dongshan Island  of Fujian Province

    Based on the field investigation,specimen sorting and literature review,composition,relationship with neighboring areas,life forms,leaf characteristics,leaf quality characteristics,ornamental characteristics,greening purpose and economic purpose of cultivated yellow flower plants in Dongshan Island of Fujian are analyzed.The results showe that there are cultivated yellow flower plants of 124 species of 87 genuses of 48 families,and the cities with more common species are mainly located the  in southern coastal region of Fujian.The cultivated yellow flower in Dongshan Island is  mainly nanophanerophytes,which belongs to 90 species,accounting for 72.58% of the total species;the leaf type is mainly of small and medium,and there are 76 species,accounting for 61.29% of the total species;the leaf quality is mainly paper and leathery,there are 79 species,accounting for 63.71% of the total species.The  flowering seasons are in spring,summer,and autumn three season,the fruiting seasons are in summer and autumn.The major types of ornamental plants includes flower and tree appreciation type,ornamental tree type and leaf appreciation type.These three ornamental types of plants have a total of 72 species,accounting for 57.11% of the total species,which can be mainly used in landscaping as flower appreciation,standing tree and street tree.The Island cultivated yellow flowers plants in Dongshan are economically widely used,there are 86 species for medicine and woody vegetables,accounting for 68.36% of the total species.

    Phylogenetic Analysis of Three Wild Grapes Based on PsbA-trnH,RbcL and trnL-F Sequences

    PsbA-trnH,RbcL and trnL-F fragments were amplified from the three wild grapes in Xiangxi Autonomous Prefecture by PCR,and cloned into pMD18-T vector for sequencing.Based on PsbA-trnH,RbcL and trnL-F gene sequences,the phylogenetic trees between these three wild grapes and other related Vitaceae plants were constructed by software Mega4 for analyzing their phylogenetic relationship.The results showed that all of these three cpDNA fragments could be used as molecular markers in phylogenetic analysis among genera of Vitaceae,but of which results had a little difference from that of traditional morphological methods.Furthermore,RbcL gene fragment could be used for molecular identification among Vitis species.

    The spaA gene encoding mature surface protective antigen A (SpaA) without signal peptide was amplified from genomic DNA of E.rhusiopathiae C43065 by PCR,The BamHⅠand HindⅢ digested PCR product was cloned into prokaryotic expression vector pET32a to generate a recombinant plasmid pET-spaA.The recombinant protein rSpaA was expressed in E.coli BL21 harboring the recombinant plasmid pET-spaA by IPTG inducing,and the expressed protein was determined by SDS-PAGE.The DNA sequence analysis showed that the spaA gene of C43065 strain was 1794 bp in length.SDS-PAGE analysis revealed a single protein band with a molecular weight of 86 kDa successfully expressed in E.coli BL21.The expressed protein of rSpaA will contribute to further study on protective domain of this protein.

    Under drought stress,bryophyta shows a series of physiological and ecological adaptation changes.This study shows bryophyta’s  physiological metabolic indexes under drought stress,and explains the relationship between desiccation tolerance and ecological adaptation.Through further analysis,bryophyta’s adaptation is shown to be controlled by many factors,so desiccation tolerance of bryophyta should be considered  comprehensively.

    Habitat and Sexual Reproductive System in Vesicularia Montagnei,a Rock Outcrop Moss in a Stream

    Based on the field investigations and laboratory examinations on V. montagnei,the authors studied its habitat and sexual reproductive system.The results were described as following:(1) the habitat of  V. montagnei is very special,which is the surfaces of rocks in a stream.Part individuals live in flooded condition constantly,but most individuals grow in the flooded condition intermittently.(2) The average number of archegonia is (18.2±6.2)(N=30) in per perichaetium,the average number of antheridia is (8.3±2.7)(N=30) in per perigonium,the average length of sporophyte is (1.8±0.17) (N=20) cm,the average length of seta is (1.6±0.16) (N=20) cm,and the average length of capsule is (0.2±0.02) (N=20) cm.(3) The development period of sporophyte in V. montagnei is between June and September among which embryo period is in June,capsule formation takes place in July,spore maturity period is concentrated on between August and September.

    In recent years,because of rapid social economic development and increasing  demand for construction land in Xiangxi Autonomous Prefecture,imbalance between the  supply of and demand for construction land has become increasingly acute.Determining a reasonable construction land scale is significant for  sustainable development of the local economy.This article analyzes the cause of expanded demand for construction land in Xiangxizhou and the mechanism of investment pressure on fixed assets using PSR model in system engineering.A PSR model for demand for construction land is established.Based on the data from 2000 to 2008,this article concludes that demand for construction land will be 3 233 hm2 in Xiangxizhou from 2008 to 2020.

    Microbial Detoxification of Hexavalent Chromium from Chromium-Containing Slag

    In this paper,treatment and recovery of hexavalent chromium(Cr(Ⅵ)) from chromium-containing slag by bacteria was studied.The work showed that a bacterial strains of Ch-1 isolated from chromium-containing slag can efficiently accelerate Cr(Ⅵ) leaching and remove it.Moreover,bacteria Ch-1 can effectively change the structure of chromium-containing slag by Scanning Electron Microscopy(SEM) and the Energy Dispersive X-Ray (EDX),which is benefitial to Cr(Ⅵ) leaching and its detoxification by changing Cr(Ⅵ) to Cr(Ⅲ).The effect of detoxification by bacteria of Ch-1 is obvious,the toxicity in leached slag was 3.3 μg/g lower than the national standard of 5 μg/g,and the recovery rates of hexavalent chromium are more than 90%.

    Influence of Habitat Inside and Outside Valley on Morphology and Cytology of Sambucus Chinensis

    Sambucus chinensis Lindl,a large herb widely distributed  inside and outside valley in northwestern Hunan province were selected for the  exploring study on the influence of on valley special-habitat cytology  and  plant morphology.(1) Morphological comparison of Sambucus chinensis  outside valley and inside valley indicates the difference in number of unfertile flowers,long/wide of leaflets,fruit color and stipule shape,but there are no obvious differences in the number of arris,the number of lateral branches,number of pitches,length of longest internodes and number of leaflets;(2)The leaf epidermal cells are usually irregular bearing with the anticlinal walls undulate;the stomatal density of S.chinensis of inside population is obviously higher than that of the outside one,37.6±7.47 per mm2 and 26.8±5.81 per mm2,respectively;while the stomatal index of S.chinensis of outside population (31.38%) is slightly higher than that of the inside one(29.17%);(3)Mitotic chromosome numbers of S.chinensis  outside and inside valley are diploidy with 2n=36 reality,and is incompatible with previous reference.Karyotype of S.chinensis is 2B and inside population with a karyotype formula of 2n=36=16m+14sm+2st+4t,and 2n=36=22m+14sm for outside population.Besides,the population inside valley appears more advanced than the outside population for their Ask values are 67.76% and 61.97%,respectively.

    The traditional knowledge and awareness of shared interests on animal epidemic disease prevention and control among local aboriginal in Xiangxi  were surveyed among farmers,livestock farmers,rural veterinarians and livestock traders through individual interviews,group discussions and questionaire from July to September in 2011.The results show that the indigenous people (aged 35~55) is the not only major carriers of traditional knowledge of livestock disease prevention and control,but also  the main executer in livestock industry;small-scale livestock farmers rely mainly on the local veterinary medicine for sick livestock during early treatment;although local indigenous people  possess very important  traditional knowledge of livestock disease prevention and control,they do not know how to protect or share the benefits of the traditional knowledge.Meanwhile,according to the properties and uses for the treatment and prevention,Local herbs are classified  into 15 categories,and  10 prescription commonly used by local indigenous for animal disease treatment are also listed.

    Influence of Seedling Culture from Different Segments of Kudzuvine Root on Seedling Growth

    The upper,middle,and lower segments of Kuzdvine root were sheaved for seedling culture.Emergence rate,average plant height,and root length and thickness being statistic index,the growth of seedlings were analyzed.The results show that the middle segments have the highest emergence rate(above 80%),and average height,length and thickness the seedlings from the middle segments are higher than those from other segments.Moreover,seedlings from the middle segments have rhizobium.In conclusion,seedling culture from middle segments of Kudzuvine root can be promoted.

    Sagra femorata is the main pest for Kudzuvine Root.The authors observed its life habit and  harm to Kudzuvine Root for three years.The results show Sagra frmorata does harm to Kudzuvine root in the whole of its life course,especially in its larva period,when the harm is the most serious and lasts the longest.In conclusion Sagra femorata does great harm to the growth and production of Kudzuvine root.

    Musk deer (Moschus spp.) are peculiar to the Palearctic and of considerable value worldwide.The high mortality-rate of fawns,especially caused by diarrhea or enteritis,has severely hindered the development of Chinese musk deer farming.In this study,two strains of pathogenic Enterobacteriaceae were isolated from the intestinal contents of an Alpine Musk Deer (Moschus chrysogaster) which died from digestive diseases at Xinglongshan Musk Deer Farm,Gansu Province,China.They were identified as strains of Escherichia albertii,based on an integrated analysis of morphological,physiological and biochemical characteristics as well as phylogenetics of genetic sequences 16SrRNA.The results of antibiotic sensitivity tests showed that the sensitivities of the bacteria to different antibiotics varied.This might have resulted from the animal’s long treatment for colibacillosis by a fixed set of antibiotics,including penicillin G and tetracycline,which might have lost efficacy because of bacterial resistance.A feasible approach to controlling colibacillosis in future would be alternative use of antibiotics combined with other preventive measures.

    The effect of starvation for different days (0,2,4,6 and 8 days) and compensatory growth on energy budget of juvenile cobia (Rachycentron canadum) was estimated by an experimental ecological method.Results in present study indicated that energy content and body weight of juvenile cobia decreased significantly after starvation,and during the re-feeding period,ingestion rate in energy and specific growth rate in energy,wet weight and dry weight in the deprived fish were higher than those of control fish.Results also indicated that juvenile cobia showed complete compensatory effect in the recovery growth and it was contributed by the increase of ingestion rate.Energy budget of juvenile cobia fed to satiation was 100.00C=38.33G+15.06F+7.22U + 39.39R,and expression by assimilated energy was 100.00A=49.32G+50.68R.

    The pigment in the browse leaves was extracted from L. Chinese Var. Rubrum leaf with Microwave treatment and distilled water or 20% EtOH.The antibacterial activities of the extracts on the three kinds  bacteria were determined by Oxford Cup method.If the extracts can abstrain the bacteria obviously,the MIC of crude drug were determined by microdilution technique.The results show that the  extracts from browse leaves by distilled water under microwave treatment have obviously antimicrobial activity on three kinds bacteria,the MIC of crude drug of the extracts from browse leaves on E. coli,shigella dysenteroae,staphylococlus aureus are 0.014 g/mL,0.010 g/mL,and 0.010 g/mL.The extracts from  browse leaves by 20% EtOH were obviously active against staphylococlus aureus and shigella dysenteroae but can not abstrain the E.coli.
