    Key Genes for Polysaccharide Synthesis Pathway from Polygonatum Based on WGCNA

    In order to find possible key genes related to the synthesis of polysaccharides in Polygonatum,this study explored the correlation between the expression patterns of core genes in leaves and rhizomes of Polygonatum cyrtonema Hua and Polygonatum odoratum (Mill.) Druce in combination with polysaccharide synthesis pathways from KEGG database using transcriptome data with expression difference and weighted gene co-expression network analysis (WGCNA).Our results indicated that FBA (fructose-bisphosphate aldolase) and GOLS (galactinol synthase) showed a strong correlation in Pearson linear regression calculation,which demonstrated that they are possible key genes for polysaccharide synthesis pathway in Polygonatum.

    N-propyl-3,4,9,10-perylene terephthalic-3,4-alcohol ester-9,10-bisimide was synthesized under ultrasonic microwave radiation with N-propyl-3,4,9,10-perylene terephthalic acid and alkyl halide,aliphatic alcohol as solvent.Results showed  that this reaction afforded corresponding compound with 65.7% yields.The optimized reaction condition was as follows:700 W of microwave power,60 min of reaction time,90℃of reaction temperature,and 80% of ultrasonic power.

    This paper simulated the spray characteristics of diesel,methanol and water emulsion by Fluent,and obtained the particle trajectory figure and particle velocity vector diagram of the spray.Meanwhile,the influence of the spray injection pressure,environmental pressure and nozzle diameter is studied on the emulsion spray characteristics.This research provides certain guiding significance for the diesel,methanol and water emulsion application.

    Factors of Influencing Gemmae Germination and Growth of Huperzia Serrata

    The factors of influencing the gemmae germination of Huperzia serrata and growth condition are explored.The effect of culture medium,culture temperature,and light intensity and GA3 on the gemmae germination condition of Huperzia was studied,and the growth morphogenesis during the course of gemmae germination of Huperzia serrata was observed at the same time.Results showed that the best culture temperature for the gemmae germination was (22±2)℃,and the germination  rate  of the optimum  medium for Moore,the germination rate was as high as 82%,and the average plant length could be up to 14.620 mm.The second culture medium for the gemmae germination was Knudson C,the germination rate was 64%,and the average adventitious root length was about 5.158 mm.Darkness was conducive to gemmae germination,the germination rate was 92%,and the average adventitious root length was about 9.088 mm.Under 800lx weak light,the adventitious root grew slowly,and the adventitious root length could be up to 8.675 mm.1600lx light irradiation exhibited some inhibition for the adventitious root growth.And GA3  for gemmae germination and growth exhibited some inhibition. This study establishes the gemmae germination culture system for Huperzia serrata,which consitutes the foundation for further study on the growth,physiological and biochemical change of Huperzia serrata.

    Characteristics Analyses of Kitchen Waste Leachate

    The kitchen wastes were collected from a student canteen in Jishou University to produce kitchen waste leachate in the laboratory in an anaerobic acidogenic reactor.The characteristics of kitchen waste leachate were analyzed.Results show that the concentration of COD and NH+4—N  for formed kitchen waste leachate after reaction for 8 weeks is more than 140.6 g/L and 16.2 g/L,respectively.With the increase of fermentation time,the pH of the kitchen waste leachate decreased from 5.9 in the end of 1st week to 3.7 in the end of 8th week.At the same time,the VFA concentration increased from 6.7 g/L to 17.9 g/L and the COD concentration increased from 57.9 g/L to 140.6 g/L,while the NH+4—N concentration increased 0.3g/L to 16.2 g/L.Prolonging the reaction time,the pH of the kitchen waste leachate tends to be stable and the concentration of VFA and NH+4—N  increases slowly,while the concentration of COD has a downward trend.

    UV spectrophotometry for determination of total flavonoids was used.Via ultrasonic extraction technique,using NaNO3-Al(NO3) to detect the absorbance of samples and taking lutin as comparison,the contents of total flavonoids were determined.On the basis of single factor experiments,the orthogonal experiment was used in the optimization of technological parameters:extracting temperature,ethanol concentration,extracting time,the ratio of solvent to raw materials.The result showed that the contents of total flavonoids in different kinds of Capsella bursa-pastoris L. were from 0.49% to 3.83%.The contents of total flavonoids in flowers & fruits were the highest,and then in leaves and stems.The contents of total flavonoids in cultivated varireties were higher than those in wile varireties.The optimum extraction conditions were as follows:temperature at 65 ℃,50% ethanol concentration,ultrasonic extraction for 40 min,and the  25∶1 ratio of solvent to raw materials.In these conditions,the total flavonoids extraction rate was 89.26%.

    Many traditional methods used to synthesize LiFePO4 (LFP for short) have the disadvantages of long reaction time or unavoidable oxidation of Fe2+.In this work,the combustion method was developed to facilely synthesize LFP by using ferrous sulfate,diammonium hydrogen phosphate and lithium nitrate as main raw materials.The synthesized LFP was characterized by XRD and TG-DSC.The results showed that the combustion method could facilely synthesize purer LFP at 600 ℃ in a short period without the protection of inert gas.The studies on combustion mechanism indicated that from 120 ℃ to 290 ℃ the organic citric acid in the dry gel was combusted which could lead to a self-propagating combustion reaction and bring in carbon preventing the oxidation of Fe2+ simultaneously.And  φ=n(CA)/n(LFP)=2 is the best amount of citric acid to synthesize LFP materials.

    In order to make full use of valuable element silicon in stone coal,alkali leaching vanadium process was used in extracting vanadium and silicon from stone coal. Through pre-roasting,the stone coal structure can be destroyed and the leaching rate of vanadium and silicon can be improved.When the roast temperature was 850 ℃,the roast time was 2 h,the leaching temperature was 95 ℃,the leaching time was 4 h,the solid ̄liquid ratio was 1∶1.4,and the ratio of alkali and ore was 1.2∶1,the leaching rates of vanadium and silicon were 86.6% and 61.4% respectively.

    This paper studies the nitrogen distribution in overlying water and sediment in the Dongguan segment of Dongjiang River.The indoor column experiment of nitrogen exchange between overlying water and sediment was conducted,and the influential factors on the nitrogen migration were investigated.The results indicated that TN concentration ranged from 2.13 to 4.48 mg/L,and nitrate was found to be the main form of contaminant,which accounted for 64.9% of TN.Sediment is an important source for ammonium and a sink for nitrate and nitrite.Illumination and disturbance are conducive to decrease the concentration of ammonia nitrogen;and higher temperature and initial pH value are conducive to the transformation of ammonia nitrogen.

    The contents of chlorogenic acid in leaves of Eucommia ulmoides Oliver from seven counties and one city in different habitats of Western Hunan were determined.The result showed that the contents of chlorogenic acid were significantly different in different habibats.The contents of chlorogenic acid were higher in the leaves of Eucommia ulmoides Oliver in the adrets of elevation  401~500 m,in light-grey soil,and with the parent material of limestone or sand shale.

    ESA/AMPS copolymers has been synthesized with MA and AMPS as raw materials and ammonium persulfate as initiator.By static scale inhibition experiment,various experimental conditions on the inhibition rate of calcium carbonate scale and calcium phosphate scale by ESA/AMPS copolymers were examined.The results show that when monomer ratio m(MA)∶m(AMPS)=3∶1,the mass fraction of initiator is 4%,and the polymerization reacts for 3 h at 85~90 ℃,scale inhibition can achieve the optimum effect.

    Storability Differences Between Yellow and Black Seeds of Xiangzayou 631

    To investigate the relationship between seed color and storability,the artificial aged Xiangzayou 631 seeds have been used as materials to study the germination and antioxidative physiological and biochemical variation.Results showed that the germination rate of both yellow and black rape seeds decreased significantly after accelerated artificial aging,and germination of yellow seeds decreased dramatically faster than the black. Superoxide radicle production,contents of hydrogen peroxide and MDA increased by 3 d accelerated artificial aging treatment,activities of antioxidative enzyme (SOD and CAT) increased obviously,and the content of AsA declined obviously.The comparison between two color seeds showed superoxide radicle production,hydrogen peroxide contents of the yellow seeds were much higher than those of the black seeds,but the SOD activity and AsA content of yellow seeds were much lower than those of the black seeds.It can be concluded that the black seeds of Xiangzayou 631 possess stronger storability due to greater contents of  nonenzymatic antioxidants.

    To investigate the effect of 16S rRNA gene and groEL gene used in the classification and identification of species of the genus Erysipelothrix,the 16S rRNA and groEL genes of four E.rhusiopathiae strains were amplified by PCR and sequenced and their phylogenetic threes was constructed.The open reading frame of groEL genes of four tested strains were 1 614 bp in length,and sequence similarities of the groEL gene between the four tested strains and the previously reported E.rhusiopathiae type strain ATCC19414 and E.tonsillarum type strain ATCC43339 were 99% and 86%,respectively.While the 16S rRNA genes of the four tested strains was 1 351 bp in length,and sequence similarity of the 16S rRNA gene between the four tested strains and E.rhusiopathiae type strain ATCC19414 and E.tonsillarum type strain ATCC43339 was 99%.Phylogenetic analysis indicated that the E.rhusiopathiae and E.tonsillarum were differentiated well by groEL gene sequence analysis.This study demonstrated that the groEL gene is an alternative phylogenetic marker for the classification of species of the genus Erysipelothrix.

    Based on the actual investigation and literature review,it is found that in western Hunan,there are 101 families,321 genera and 101 species (including the cultivation of 8 families,29 genera and 76 species) of forest resources,which can be roughly divided into four categories of timber forests,gardening forests,economic forests,and firewood forests.Chinese fir occupies an important position in the western Hunan forest resources,with a total of about 10 kinds of 6 genera and 3 families being collectively referred to as “Chinese Fir”,which is not only known as “omnipotent woods”,but also has specific cultural meanings.Forest resources in Xiangxi region are significant in three aspects of the forestry economic,ecological maintenance and forest culture.

    The genes encoding the outer membrane protein H (OmpH),lipoprotein E (PlpE),type 4 fimbrial subunit protein (PtfA) and fibronectin binding protein (PM1665) were amplified by PCR from genomic DNA of avian Pasteurella multocida P-1059,respectively.The PCR products were cloned into the pMD18-T vector and then sequenced.Sequence analyses showed that the open reading frames (ORFs) of the ompH,plpE,ptfA and pm1665 genes were 1 032,1 008,435 and 348 bp in length,and encoded the 343,335,144 and 115 amino acid residues.Nucleotide sequence homologies of the ompH,plpE,ptfA and pm1665 genes between the P-1059 and the previously reported different serotype strains of P. multocida were 95%~97%,95%~99%,94%~97% and 99%,respectively.In conclusion,the cloned genes encoding the adhesive proteins of avian P. multocida P-1059 will contribute to further study on pathogenesis of this organism.

    The field experiment and instrument analysis method to study the trace element selenium compound fertilizer influence and the yield and selenium content of peanut in Hepu of Beihai.The results showed that:in the flowering period and flowering period of peanuts,sprayed the Selenium enriched fertilizer,could significantly increase the content of peanut selenium.Selenium content increased with the frequency of spraying,the selenium content is 1.031 μg/g,improved more than times of conventional control.In the meanwhile,the yields was increased six percent.It can make the selenium content in selenium enriched standard to peanut by sprayed Selenium enriched fertilizer.

    Different Expressions of vscO Gene from Vibrio Alginolyticus in Different Environments

    To understand the impact of environmental factors on the expressions of vscO gene,the mRNA expressions of vscO gene in different growth periods and with environmental factors,such as temperature,salinity and pH,were detected by RT-PCR method. The results indicated that the mRNA expression of vscO reached a maximum at 28 ℃ in salinity of 2%,with the initial medium pH 7.0 when V. alginolyticus was in the middle phase of growth.
