Abstract: Using Polygonatum odoratum (Mill.) Druce powder as the raw material, this study investigated the effects of  chloroform to butyl alcohol volume ratio,Sevage reagent to polysaccharide sample volume ratio, and shaking time on polysaccharide retention rate and protein removal rate.Based on single factor experiments, the protein removal process for polysaccharides was optimized using response surface methodology. The optimal conditions were determined as follows:V(chloroform)∶V(butyl alcohol)=4∶1,V(Sevage reagent)∶V(Polysaccharide  sample)= 1∶2,and shaking time of 30 min.Under these conditions, the polysaccharide retention rate reached  76.01%,and the protein removal rate was 28.73%.This optimized protocol provides a reliable approach for efficiently removing proteins while preserving polysaccharide integrity during the purification of Polygonatum odoratum (Mill.) Druce polysaccharides.
