Screening and Identification of a High-Temperature Resistant Cellulose-Degrading Bacterium

Abstract:

In order to obtain high-temperature resistant cellulose-degrading bacteria,samples were collected from chicken manure and mushroom residue compost.After being enriched through filter paper medium,high-temperature resistant cellulose-degrading bacteria were isolated and purified on cellulose Congo red medium.The isolates were preliminarily screened through filter medium and cellulose Congo red medium,and then were further screened through quantitative analysis of cellulase activity of liquid fermentation broth.A high-temperature resistant cellulose-degrading bacterium,designated strain XD-3,was obtained,which grew well at 50 ℃ and the carboxymethyl-cellulase (CMCase) activity was 6.14±0.37 U/mL.The combination of morphology,physiological/biochemical characterization and phylogenetic analysis based on 16S rRNA gene sequence showed that strain XD-3 was a member of a known Bacillus species,Bacillus amyloliquefaciens.
