Content Measurement of Oleanolic Acid in Akebia Trifoliata Pericarp by Ultraviolet-Visible Spectrophotometry

Abstract:

In order to establish a spectrographic method of measuring oleanolic acid,conditions of UV-spectrophotometry analysis was investigated with vanillin-glacial acetic acid as chromogenic reagent and 545 nm as the determination wavelength.The results showed that the measured concentration value of 8.00 mg/L oleanolic acid standard solution was 8.02 mg/L with 0.3 mL vanillin-glacial acetic acid as chromogenic agent,duration was 15 min under 70 ℃,linear relation of the oleanolic acid was good in 2.00~16.00 mg/L,and the correlation coefficientwas 0.999 8.Content of oleanolic acid in Akebia trifoliata pericarp was 3.70 mg/g by this analysis method.Standard addition recovery rate was 101.06%,and its relative standard deviation (RSD) was 1.22%.RSD of precision,repeatability and stability was 0.02%,0.29% and 1.29%,respectively.
