The Effect of Salicylic Acid on Expression of the Gene Coding Glycine-Rich Protein Binding RNA in Tobacco

Abstract: The gene fragments(GRPs) coding glycine-rich protein binding RNA in tobacco materials were got by RT-PCR.Northern hybridization blot analysis was used to demonstrate the expression pattern of the GRP gene at different times after treatment of salicylic acid in different tobacco materials.Inducded by salicylic acid,the GRP genes of Nicotiana could up-regulate expression.The expression attained the highest in 3h,then began descending.The three tobacco varieties had different resistance abilities to TMV.The expression level of two genes correlated with the resistance ability.The increase of two genes in Honghuadajingyuan with stronger resistance ability was higher than that of Yunyan 85 with less resistance ability.The expression pattern of the gene induced by salicylic acid in tobacco was got.It was found that in different tobacco varieties with different resistance abilities,the up-regulation extents of expression of the GRP gene were quite different after treatment of salicylic acid.And it is also found that the up expression became significant after 3 hours treatment.From the results of this study,it is suggested that GRP gene can be used as a marker gene to investigate salicylic acid metabolism in tobacco varieties so as to demonstrate their response to abiotic stresses.
