To study the mechanism of Caulis Sinomenii in the treatment of rheumatoid arthritis, network pharmacology and molecular docking were used to screen 6 main active ingredients of Caulis Sinomenii from the TCMSP database.268 drug targets corresponding to the active ingredients were obtained through the PubChem library and Swiss Target Prediction.Subsequently,5 706 targets for rheumatoid arthritis were obtained through GeneCards,DisGeNET,and OMIM databases.By utilizing the Venny 2.1.0 platform to draw Venn diagrams,178 intersection targets between the active ingredients of Caulis Sinomenii and rheumatoid arthritis diseases were identified.A network diagram illustrating the interaction between the active ingredients of Caulis Sinomenii and rheumatoid arthritis disease was constructed using Cytoscape 3.10.0.Additionally,a drug component target protein network diagram was created.GO enrichment analysis and KEGG pathway enrichment analysis were conducted using the DAVID database.Molecular docking simulation was performed between the target and the active ingredients of the drug using Autodock software. The results revealed that NR3C1,AKT1,SRC,PPARG,and JUN are the key target proteins for the anti-rheumatoid arthritis effect of Caulis Sinomenii. The GO enrichment analysis results indicated that biological processes mainly include G protein-coupled receptor signaling pathway, coupled to cyclic nucleotide second messenger G,vascular process in circulatory system, adenylate cyclase-modulating G protein-coupled receptor,peptidyl-serine phosphorylation, etc. The cellular components mainly include integral component of presynaptic membrane, intrinsic component of presynaptic membrane, membrane raft, membrane microdomain, etc. The molecular functions mainly involve nuclear receptor activity,ligand-activated transcription factor activity, catecholamine binding, protein tyrosine kinase activity, etc.According to the KEGG enriched bubble diagram,it can be inferred that the affected pathways include the cancer pathway, the neuroactive ligand receptor interaction pathway, and the inflammatory mediator regulatory pathway of the TRP channel. The molecular docking results indicated that the active ingredients, including β-sitosterol,razmanine,xylosterol,sinomenine,and levodontidine, are associated with the core targets NR3C1,AKT1,SRC,PPARG,and JUN,all showing good binding activity. The treatment of rheumatoid arthritis with Caulis Sinomenii may involve a multi-component,multi-target,and multi-pathway process, which includes regulating inflammatory cytokines, immune cells, cell cycles (such as apoptosis and burning),and osteoblasts and osteoclasts to treat rheumatoid arthritis.

A simplified one-dimensional thin-layer chromatography (TLC) method has been established for the identification of Paulownia species leaves, including Paulownia fortunei,Paulownia elongata,Paulownia tomentosa and Paulownia kawakamii.A single-factor experiment was conducted to select the optimal two-dimensional multiple development TLC method, which was then combined with column chromatography to segment the extract. Then, the one-dimensional TLC method was employed to identify the leaves of the four Paulownia species after developing the concentrated segments. The results showed that in the two-dimensional multiple expansion, characteristic spots of Paulownia fortunei were identified.Additionally,in the one-dimensional thin layer after segmented concentration, four common characteristic spots were observed for the four Paulownia species at 254 nm,seven common spots at 365 nm,and twelve common spots after color reaction under sunlight. The differences between Paulownia fortunei and the other three species were noted:on the developed TLC plate after color reaction, the first segment of the small polar sample showed a slightly yellow spot at an Rf value of 0.5,while the third segment of the large polar sample exhibited two light pink spots at an Rf value of approximately 0~0.2.

A tail suspension analysis system has been established based on DeepLabCut (DLC) algorithm for evaluating depressive behavior in mice.Based on the DLC algorithm in deep learning technology,multi-path,semi-closed tail suspension hardware, the systems of software and hardware were constructed;this system and open field analysis system were applied to evaluate the tail suspension and open field behavioral changes in mice with reserpine depression model, intermittent alcohol consumption injury model, and periodontal silk ligation model. This system realized the synchronous analysis of head movement and limb movement in tail suspension, and automatically quantified 50 tail suspension indexes of dynamic and static type, posture type, and intensity type.It was found that in the reserpine depression model, compared with the control group, the movement distance, wall climbing times, and head swing times were reduced in the low-,medium-,and high-dose reserpine groups in 2 days, and the head swing times were reduced in the medium- and high-dose reserpine groups in 3 days and 4 days.In the intermittent alcohol consumption injury model, compared with the control group, the immobility time was reduced, the limb struggling times, movement distance, and movement time were increased in the low- and medium-dose ethanol groups, and the forelimb, hindlimb, and absolute struggling times were reduced in the high-dose ethanol group. In the periodontal silk ligation model, the movement distance and length, limb and absolute struggling times, and the total value of forelimb, hindlimb, and limb angles were reduced in the ligation group.Clearly, the DLC algorithm-based mouse tail suspension analysis system can accurately distinguish the behavioral characteristics of different mechanistic depression models.
