    GC-MS Fingerprint of Petroleum Ether Fraction in Paulownia fortunei Leaves

    To evaluate the quality of Paulownia fortunei leaves from different regions,gas chromatography-mass spectrometry was used to qualitatively analyze the petroleum ether fraction of Paulownia fortunei leaves.Squalene used as a control,the similarity evaluation system of traditional Chinese medicine chromatographic fingerprint (2012A version) used for similarity evaluation,cluster analysis and principal component analysis were combined to evaluate the quality.The results indicated that a total of 12 common peaks were identified in 19 batches of Paulownia fortunei leaves from various regions.These peaks corresponded to the following compounds:4,4,7a-Trimethyl-5,6,7,7a-tetrahydro-1-benzofuran-2(4H)-one,phytol,heptadecanoate,2-methyl octacosane,squalene,nonacosane,triacontane,hexatriacontane,α-Tocopherol,nonatriacontane,tetracontane,β-Sitosterol.Through cluster analysis,19 batches of Paulownia fortunei leaves were divided into 3 major categories.The cumulative variance contribution rate of the two principal components obtained from principal component analysis was 86.725%,and the results showed that sample S12 had a higher comprehensive score.The results of principal component analysis and cluster analysis were basically consistent.

    Application of Simplified Thin-Layer Chromatography in the Analysis of Paulownia fortunei Leaves

    A simplified one-dimensional thin-layer chromatography (TLC) method has been established for the identification of Paulownia species leaves,including Paulownia fortunei,Paulownia elongata,Paulownia tomentosa and Paulownia kawakamii.A single-factor experiment was conducted to select the optimal two-dimensional multiple development TLC method,which was then combined with column chromatography to segment the extract.Then,the one-dimensional TLC method was employed to identify the leaves of the four Paulownia species after developing the concentrated segments.The results showed that in the two-dimensional multiple expansion,characteristic spots of Paulownia fortunei were identified.Additionally,in the one-dimensional thin layer after segmented concentration,four common characteristic spots were observed for the four Paulownia species at 254 nm,seven common spots at 365 nm,and twelve common spots after color reaction under sunlight.The differences between Paulownia fortunei and the other three species were noted:on the developed TLC plate after color reaction,the first segment of the small polar sample showed a slightly yellow spot at an Rf value of 0.5,while the third segment of the large polar sample exhibited two light pink spots at an Rf value of approximately 0~0.2.

    Thin-Layer Chromatography Identification of Paris fargesii Rhizome,Trillium tschonoskii Rhizome,and Paris Rhizome

    Paris Saponin II,VI,H were used as reference substances,and the rhizomes of Paris fargesii,Trillium tschonoskii,and Paris (The rhizome of Paris polyphylla Smith var. chinensis (Franch.) Hara and Paris polyphylla Smith var. yunnanensis (Franch.) Hand.-Mazz.) were identified by thin-layer chromatography (TLC).The results showed that the three reference substances had high resolution and clear spots on the silica gel G thin-layer plate with chloroform-methanol-water (15∶5∶1) as the developing agent under the conditions of 20 ℃±2 ℃ and 50%~60%.According to the difference in components,TLC can be used to distinguish Paridis fargesii rhizome,Trillium tschonoskii rhizome,and Paris rhizome.
