Protective Effect of Hydrogen Sulfide on the Injury of Endothelial Cell Induced by Oxidized HDL

摘要:探讨硫化氢对氧化高密度脂蛋白(ox-HDL)致人脐静脉内皮细胞(HUVECs)损伤的保护作用.方法 体外培养HUVECs细胞,应用形态学观察法和噻唑兰(MTT)比色法检测细胞活性,试剂盒测定培养液中乳酸脱氢酶(LDH) 及丙二醛(MDA)含量.结果 ox-HDL组细胞损伤明显,经硫化氢低、中、高剂量组的干预,受损情况均有较明显改善,细胞存活率增加,且培养液中LDH及MDA含量降低,与对照组比较差异有显著性(P<0.05).结论 硫化氢对ox-HDL所致内皮细胞损伤具有一定的保护作用,其机制可能与降低氧自由基抗氧化作用有关.

Abstract: Objective To investigate the protective effect of hydrogen sulfide on the injury of endothelial cell induced by ox-HDL.Methods HUVECs were incubated in vitro.The cells viability is measured by morphological observation and the MTT assay.The levels of LDH and MDA activity in culture medium were measured with corresponding kit.Results HUVECs were damaged obviously in ox-HDL group,hydrogen sulfide improved the condition of HUVEC significantly,increased the cell survival rate,decreased the level of LDH and MDA(P<0.05).Conclusion These results demonstrate that hydrogen sulfide can protect HUVECs from damage by ox-HDL,and it may be through decreasing oxygen free radical production and antioxidation.
