Flavonoid Compounds Separation with Coagulation Activity from Artemisia annua L.by Liquid-Liquid Chromatography

Abstract: A liquid-liquid chromatography method based on a spiral separation tube was developed for the separation and purification of anthelmintic flavonoids from Artemisia annua L..To achieve the best separation effect,the mobile phase system and elution flow rate were systematically optimized,and HPLC analysis was used to further verify the separation effect.The results showed that the optimal mobile phase for the separated flavonoids from pH7,pH6,pH5,and pH4 samples was petroleum ether,petroleum ether-ethyl acetate (V(petroleum ether)∶V(ethyl acetate)=3∶1),petroleum ether-ethyl acetate (V(petroleum ether)∶V(ethyl acetate)=3∶1),and petroleum ether-ethyl acetate (V(petroleum ether)∶V(ethyl  acetate)=1∶8),respectively;the optimal flow rate was 3 mL/min;after liquid-liquid chromatography separation of the flavonoids from pH7,pH6,and pH5 samples,a single component could be achieved;after separation of the flavonoids from pH4 sample,there were 4 components in the elution flow.It is suggested that the liquid-liquid chromatography based on the spiral separation tube can effectively separate the anthelmintic flavonoids from Artemisia annua L..
