湘西杜鹃花根和茎中氨基酸含量测定

Analysis of the Amino Acids in the Root and Stem of Rhododendron Simsii Planch by HPLC Method

摘要:用 HPLC 方法对湘西杜鹃花植物根、茎中的15 种氨基酸的含量进行了测定. 测定的结果是, 根中氨基酸的质量分数为 12.74 ‰ ( 干样) ,其中必需氨基酸为 5.30‰, 占41.60%;茎中氨基酸的质量分数为8.41‰( 干样) , 其中必需氨基酸为3.25‰, 占41.85‰.该方法检测效果好, 平均回收率达 93.6%,氨基酸保留时间变异系数平均为 1.0 ±0.4%, 其峰面积变异系数为4.2 ±0.8%, 在 10 pmol ~ 2 nmol 范围内,线性关系系数平均为0.987±0.009.

Abstract: In the present experiment, fifteen amino acids in the root and stem of Rhododendron Simsii Planch( dry sample) were measured with o- phthalic aldehyde- -mercaptoethanol precolum denivalization, reversed- phase gradient elution and fluoresence detection.There are 12. 74  amino acids in the root, and 5.30  of them are essential amino acids.There are 8.41 amino acids in the stem, and 3.52  of them are essential amino acids.The satisfactory separation was obtained by this analyt ical method. The mean recovery could reach 93.6%, the mean Vaniation coefficient of retention time of fif teen amino acids was 1.0 ± 0.4% ( mean ± S. D) , while that of peak area was 4.2±0.8% , in the range of 10 pmol to 2 nmol, the amounts were linearly related with the peak areas( r=0. 987±0. 009, mean ±S. D) .
