莱菔子油抗冻伤作用及机制探索

Study on the Effect and Mechanism of Semen Raphani Oil in the Treatment of Chilblains

摘要:为了探讨莱菔子油抗冻伤的作用及机制,通过建立小鼠冻伤模型,观察莱菔子油对小鼠足肿胀率、疼痛阈值和冻伤组织病理变化的影响,并利用网络药理学技术对莱菔子油中的抗冻伤成分、靶点和信号通路进行了分析.动物实验结果表明,莱菔子油可明显缓解小鼠足肿胀,提高冻伤小鼠疼痛阈值,缓解皮肤水肿,减少冻伤组织中肥大细胞数目.网络药理学预测结果表明,莱菔子油治疗冻伤存在38个潜在靶点,MAOB,HSP90AB1,PTGS2,PTGS1,ACTB,MYC,PGR,NCOA2,RELA和RXRA靶点为莱菔子油治疗冻伤的核心靶点,主要涉及的信号通路包括神经活性配体信号、化学致癌作用-受体激活、人巨细胞病毒感染和磷脂酰肌醇-3 -激酶-蛋白激酶B信号通路等,主要涉及的生物功能包括平滑肌收缩、异生物质刺激反应和产热反应等.

Abstract: To investigate the anti-chilblain effect and mechanism of Semen Raphani oil,a mouse chilblains model was established to observe the effects of Semen Raphani oil on paw swelling rate,pain threshold,and pathological changes in chilblained tissues.Network pharmacology was used to explore the anti-chilblain components,targets,and signaling pathways of Semen Raphani oil.Animal experiments showed that Semen Raphani oil significantly alleviated paw swelling,increased the pain threshold in chilblained mice,relieved skin edema,and reduced the number of mast cells in chilblained tissues.Network pharmacology predictions indicate that Semen Raphani oil may interact with 38 potential targets in the treatment of chilblains.The core targets include MAOB,HSP90AB1,PTGS2,PTGS1,ACTB,MYC,PGR,NCOA2,RELA,and RXRA.The primary signaling pathways involved are neuroactive ligand signaling,chemical carcinogenesis-receptor activation,human cytomegalovirus infection,and the phosphatidylinositol-3-kinase-protein kinase B (PI3K-Akt) pathway.Key biological functions associated with these targets include smooth muscle contraction,xenobiotic stimulus response,and thermogenesis.
