鳞柄白毒鹅膏菌中α-amanitin的分离与鉴定

Study on the Separation and Distribution ofα-amanitin in Amanita virosa

摘要:利用反相高效液相色谱和紫外吸收光谱技术,以乙腈-0.02 mol/L醋酸铵为流动相的梯度洗脱模式,对采集自四川省阿坝州三江自然保护区的鳞柄白毒鹅膏菌子实体中α-鹅膏毒肽含量的分布情况进行研究.研究发现,α-鹅膏毒肽水溶液稳定性很好,其定量分析的线性回归方程为Y=5 159x+1 097.4,鳞柄白毒鹅膏菌子实体中α-鹅膏毒肽的含量为2 838.8 μg/g干子实体.α-鹅膏毒肽在子实体中的分布具有组织特异性,在菌裙中含量最高,约占α-鹅膏毒肽总量的51.09%,且在单位组分中,α-鹅膏毒肽的量也最高约至菌裙组干重的0.29%.

Abstract: The compound α-amanitin from Amanita virosa was separated and determined by reversed-phase high performance liquid chromatography and UV-spectrum,by using 0.02 mol/L aqueous ammonium acetate-acetonitrile as mobile phase with gradient elution mode.The concentration and distribution of α-amanitin in A.virosa carpophores were studied,which were collected in SanJiang nature reserve of ABa zhou in SiChuan province.The results showed that α-amanitin aqueous solution was stable and the linear equation of α-amanitin quantitative analysis was Y=5 159x+1 097.4.The content of α-amanitin reached about 2 838.8 μg per gram dry fruit bodies and the distribution of α-amanitin was tissue dependent.The content of α-amanitin in gills was the highest,which was about 51.09% of total toxin per gram dry carpophores,and the concentration,the 0.29% of dry gills,was also the highest among the different tissues.
